인간을 창조하고, 인간에게 능력을 주고, 인간의 목숨을 앗아가는 신같은 존재가 현미경으로 보일까 말까 하는 미토콘드리아라고 하면 믿을 수 있겠는가? 14억년 전, 박테리아였던 미토콘드리아가 다른 박테리아와 결합한 덕에 지구의 생물은 박테리아를 넘어 지금 우리가 볼 수 있는 진핵생물로 진화할 수 있었다. 박테리아는 구조적으로 크기가 제한돼있지만 진핵생물은 엄청난 에너지를 뽑아내는 미토콘드리아 덕분에 훨씬 더 크고 육식적인 세포로 진화할 수 있었고, 더 나아가 다세포 생물로 진화할 수 있었던 것이다.


미토콘드리아는 양성자구동력을 통해 에너지를 생산한다. 미토콘드리아는 내막과 외막이 있는데, 수소 원자에서 분리된 전자가 호흡 연쇠를 타고 내려가면서 애너지를 방출하고, 이 에너지로 내막 안에 있는 양성자를 내막 밖으로 밀어내고, 이 양성자들이 농도 기울기 때문에 다시 내막 안으로 들어오면서 열을 발산하거나 에너지의 단위인 ATP를 생산한다. 호흡작용이라고 불리는 이 과정은 매우 효율적으로 에너지를 생산하는데, 박테리아는 미토콘드리아와 같이 다수의 막이 없기 때문에 호흡작용을 하지 못하고, 진화가 제한된 것이다.


아이러니한것은 미토콘드리아가 생명의 다양성을 선사한 대신 생명을 앗아간다는 것이다. 위에 언급한 호흡 연쇠에서 차질이 생겨 전자의 흐름이 막힐 때 주변에 산소랑 반응해서 유리기 (free radicals)를 생산한다. 이 유리기가 미토콘드리아의 유전자에 돌연변이를 일으켜 손상되게 하고, 손상이 오랜 시간에 걸쳐 쌓이면 결국 미토콘드리아는 세포와 같이 죽는 것이다.


결국 우리가 죽는 이유는 미토콘드리아가 유리기를 만들기 때문이다. 따라서 반대로 생각하면 유리기 생산을 방지하면 더 오래 살 수 있는 것이다. 실제로 조류는 매우 적은 유리기를 생산하기 때문에 비슷한 크기의 포유류에 비해서 훨씬 오래 산다 (생쥐의 수명은 3년, 앵무세의 수명은 100년). 그 이유는 새가 날기 위해서는 극도로 많은 에너지가 필요한데, 그러기 위해선 많은 미토콘드리아가 필요하기 때문이다. 미토콘드리아가 많다는 것은 운동하고 있지 않을 때 각 미토콘드리아에 있는 호흡 연쇠에 과부하가 걸리지 않고, 따라서 유리기가 거의 생산되지 않는다.


위에 원리에 따르면 요즘 유행하고 있는 1일 1식도 과학적 근거가 있다. 섭취하는 영양분이 적으면 호흡연쇠에 과부하가 걸리지 않기 때문에 유리기가 생산되지 않는 것이다. 더 나아가 연구만 한다면 인간도 유전자 조작을 통해서 조류처럼 오래 사는 것이 가능할 것이다. 다만, 우리 자신을 실험쥐로 삼아 조작하는 것이 과연 안전하고 도덕적으로 옳은지에 대한 생각이 필요할 것이다. 인공적으로 새로운 “종”이 된다는 생각은 다소 껄끄롭지 않을 수 없다.


Would you believe it if you were told a God-like being that created us, invested us with power and take our lives is nothing more than mitochondria, a microscopic cell organelle? The world is full of diverse eukaryotes (the type of cells that animals and plants are composed of) because 1.4 billion years ago, mitochondria, which were bacteria, fused with another bacteria to form a whole new kind of organism. Unlike bacteria, whose size is limited by its structural features, eukaryotes could evolve into much bigger and carnivorous cells thanks to the mitochondria that produce huge amount of energy, and in turn could evolve into multicellular organisms.


Mitochondria produce energy using a principle called "proton motive force." A mitochondrion has an inner membrane and an outer membrance, and energy is produced when an electron that has been separated from a hydrogen atom travels down the respiratory chain, and this energy is used to push out the protons inside the inner membrane; as the protons re-enter the inner membrane down a concentration, they generate heat or produce ATP, the unit of energy in living organisms. Since bacteria don't have multiple membranes like the mitochondria, it cannot undergo respiration, which is a very efficient way of producing energy; this meant that the evolution of bacteria was limited.


The irony is that although mitochondria gave incredible diversity to life, it also takes life. If anything goes wrong and the flow of electrons is blocked in the respiratory chain, the electrons instead react with oxygen around it to form "free radicals." The free radicals damage the DNA of the mitochondrion by mutating it, and the damage that builds up over time kills the mitochondrion, killing the cell in turn.


It seems, therefore, that the reason that we age and die is because mitochondria produce free radicals. If we think the other way round, reducing the production of free radicals will allow us to live longer. Indeed, the birds produce very little free radicals, so live a lot longer that mammals of similar size (rats live 3 years while parrots live 100 years). The reason is that birds need a burst of instantaneous energy when they fly, and this requires a huge number of mitochondria. A lot of mitochondria means a lot of spare capacity when the bird is not flying, which would prevent an overload on the respiratory chains in the mitochondria, leading to limited production of free radicals.


Following the principle above, the recent trend of "one meal a day" has scientific basis. With less nutrient intake, there is less overload on the respiratory chains, so that there are less free radicals produces. Taking things further, it may even be possible to live longer like the birds do through genetic modification, as long as intensive research takes place on the topic. However, we should take a moment to consider whether treating ourselves as lab rats subject to genetic modification is safe and ethical. The idea of becoming a new "species" through artificial manipulation is not a comforting thought.
